In this study, two experiments were performed to explore the function of silymarin in adipogenesis in grass carp (Ctenopharyngodon idellus) using in vitro and in vivo models. In experiment 1, differentiated grass carp pre-adipocytes were treated with silymarin for 6 days. Treatment with 100 μg mL?1silymarin (SM100 group) significantly reduced triglyceride accumulation at day 6. The adipogenic gene expression levels of PPARγ, C/EBPα, SREBP1c, FAS, SCD1, and LPL, and the protein expression level of PPARγ were significantly down-regulated in the SM100 group. Additionally, the SM100 group had significantly lower reactive oxygen species production and reduced glutathione contents compared with the control in vitro. In experiment 2, the juvenile grass carp (mean body weight= 27.4 ± 0.17 g) were fed six isonitrogenous and isocaloric diets in a factorial design containing 0, 100, or 200 mg kg?1 silymarin (SM0, SM100, SM200) associated with either 4 or 8% lipid levels (low lipid, LL, and high lipid, HL, respectively) for 82 days. The results demonstrated that dietary silymarin supplementation significantly reduced the elevated intraperitoneal fat index in grass carp fed with high-lipid diets, and the gene expression of adipogenesis (PPARγ, FAS) when supplemented with dietary silymarin was notably lower than when no silymarin was supplemented under the high-lipid diets. Thus, our data suggest that silymarin suppressed lipid accumulation in grass carp both in vitro and in vivo, and the effect might be due to an influence on the expression of adipogenesis factors and ROS production partly associated with effects on antioxidant capability. 相似文献

Two 8‐week growth trials were conducted in indoor recirculation system to evaluate the protein requirements for juvenile (3.70 ± 0.20 g) and pre‐adult (85.2 ± 0.70 g) gibel carp, Carassius auratus gibelio var. CAS III. Six isoenergetic diets were formulated for each trial using fish meal and casein as protein sources, and protein level was 250–450 g kg?1 in Trial 1 and 200–450 g kg?1 in Trial 2. With the increasing dietary protein, feeding rate (FR) and feed conversion ratio (FCR) significantly decreased (P < 0.05). Weight gain (WG) increased first and then reached a plateau in 330–450 g kg?1 in Trial 1 (P > 0.05), while decreased after the maximum value in 350 g kg?1 in Trial 2 (P < 0.05). Productive protein values (PPVs) were lower in 370–450 g kg?1 in Trial 1 and 400–450 g kg?1 in Trial 2 (P < 0.05). Increasing dietary protein level increased protein content and decreased lipid content in whole fish body and white muscle (P < 0.05). Apparent digestibility coefficient of dry matters (ADCd) decreased, while apparent digestibility coefficient of protein (ADCp) increased in 370–450 g kg?1 in Trial 1 and 250–450 g kg?1 in Trial 2 (P < 0.05). Trypsin activity significantly increased in 370–450 g kg?1 in Trial 1 (P < 0.05) and was not affected in Trial 2 (P > 0.05). Hepatic alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities in both trials increased when dietary protein was above 400 g kg?1 (P < 0.05). Based on quadratic regression of WG, it was estimated that dietary protein requirement for maximum growth was 414 g kg?1 (digestible protein of 376 g kg?1) and 365 g kg?1 (digestible protein of 324 g kg?1) for juvenile (3.70 g) and pre‐adult gibel carp (85.2 g). 相似文献

Seed longevity is a very important characteristic controlling seed quality. However, the mechanism underlying this characteristic is poorly understood. In this study, 'FH7185', a storable rice variety, the germination rate of which was 66.63%, much higher than the control after artificial ageing under relative humidity 88% and 42°C for 21 days. Different methods were applied to reveal the involvement of proteins during ageing of rice seeds. In total, 35 differential proteins were identified from 2D‐PAGE, and 3,719 proteins from iTRAQ analysis. A comparison of these two methods showed that not all protein types could be detected on the 2D‐PAGE gels, and the dynamic range was somewhat limited. So the comprehensive proteome map from the iTRAQ analysis was used to identify the quantitatively regulated proteins that played roles in seed longevity, and found that the most marked change was the increased abundance of many metabolic enzymes, especially the ones involved in α‐linolenic acid metabolism in the embryos during ageing. OsLOX2 and OsLOX3 had a negative effect on seed longevity, which were lower in FH7185 than the control. The dehydrogenase (LOC_Os07g44430) which played a major catalytic role in lipid peroxidation, also changed significantly during ageing. Therefore, OsLOX2, OsLOX3 and Loc_OS 07G44430 may be involved in the regulation of seed longevity with a synergistic effect. 相似文献

The present study was conducted to investigate the effects of sodium butyrate (SB) on the physical barrier function, pro‐inflammatory response and possible underlying mechanisms in the distal intestine (DI) of yellow drum when fed a high‐SBM diet. Three iso‐proteic and iso‐lipidic diets were formulated with fish meal (FM, the control), 45% fish meal protein replaced by SBM (SBM) and 45% fish meal protein replaced by SBM but supplemented with 0.15% SB (SBM + SB). Fish were fed twice daily for 10 weeks. Results indicated that SB supplementation significantly increased the specific growth rate (SGR) and feed efficiency ratio (FER) and methionine content of muscle when compared with those of fish fed the SBM diet (p < .05). The morphologic histology results showed that SB dramatically improved the physical barrier structure, characterized as increases of fold height (FH) and muscular thickness (MT) (p < .05). RT‐qPCR data were accordant with morphologic histology results, in which claudin 3, claudin 4 and occludin were increased while claudin 7 and myosin light chain kinase (MLCK) mRNA expression levels were decreased (p < .05). Sodium butyrate also exerted anti‐inflammatory function, which may be attributed to the suppression of nucleus p65 protein expression. Results suggest SB can be incorporated in high‐SBM diets to ameliorate the negative consequences of alternative dietary ingredients on yellow drum physiology. 相似文献

Non-infectious prenatal mortality severely affects the porcine industry, with pathological placentation as a likely key reason. Previous studies have demonstrated that peroxisome proliferator-activated receptor gamma (PPARγ) deficiency causes defects in the uteroplacental vasculature and induces embryonic losses in mice. However, its role in porcine placental angiogenesis remains unclear. In the present study, PPARγ expression was investigated in porcine uteroplacental tissues at gestational day (GD) 25, GD40 and GD70 via quantitative polymerase chain reaction (qPCR), Western blot and immunohistochemistry (IHC). Moreover, the roles of PPARγ in porcine placental angiogenesis were investigated using a cell model of porcine umbilical vein endothelial cells (PUVECs) to conduct proliferation, migration and tube formation assays in vitro and a mouse xenograft model to assess capillary formation in vivo. The results showed that PPARγ was mainly located in the glandular epithelium, trophoblast, amniotic chorion epithelium and vascular endothelium, as indicated by the higher expression levels at GD25 and GD40 than at GD70 in endometrium and by higher expression levels at GD40 and GD70 than at GD25 in placenta. Moreover, PPARγ expression was significantly downregulated in placenta with dead foetus. In PUVECs, knocking out PPARγ significantly inhibited proliferation, migration and tube formation in vitro and inhibited capillary formation in mouse xenografts in vivo by blocking S-phase, promoting apoptosis and downregulating the angiogenic factors of VEGF and its receptors. Overall, the spatiotemporal heterogeneity of PPARγ expression in porcine uteroplacental tissue suggests its vital role in endometrial remodelling and placental angiogenesis, and PPARγ regulates placental angiogenesis through VEGF-mediated signalling. 相似文献

本文以1047年发生在北宋宫廷内的一场祈雨祭祀为研究对象,运用"剧场国家"理论,描绘了祈雨的"展演"相与"权力"相,研究了祈雨期内以展演为表征、以权力为暗流的人神、人地、人人关系。研究发现:一方面,祈雨是充满象征意义的展演式活动,祈雨人宋仁宗需要向天神与黎民展演德行,而展演德行又需借助于气象技术;另一方面,贾昌朝被罢相是多重因素作用下的结果,罢相反映出北宋中期朝廷内激烈的政治纷争,其仍为庆历新政后权力博弈的延续。可见这场祈雨是一场德行的展演,也是实在的操纵帝国的权力之争。 相似文献

为获得丰富的艾纳香内生真菌资源,筛选出抗性生防菌株,用于活性次生代谢产物的发现,通过水琼脂法对海南产艾纳香进行内生真菌分离,通过ITS序列分析,对其进行鉴定和分析。采用滤纸片法评价内生真菌发酵产物对细菌的抑制活性,采用平板对峙培养法评价艾纳香内生真菌对植物炭疽菌的拮抗作用。本研究共分离鉴定出191株内生真菌,分属于27属,45种,其中优势菌属为Diaporthe、Paraphoma、Ectophoma、Penicillium、Talaromyces、Hypoxylon、Phomopsis、Colletotrichum和Fusarium。抗细菌活性结果显示,Clonostachys、Fusarium和Diapothe属活性较强。抗炭疽病菌试验显示,Talaromyces、Ectophoma和Penicillium属活性较强。本研究初步探讨了海南产艾纳香内生真菌种类的多样性,并首次对其进行了抗病原菌活性筛查,获得的活性菌株可作为微生物源菌剂开发的重要材料。 相似文献
